Silk contains about 3/4 of silk fibroin and 1/4 of sericin. In addition to silk fibroin and sericin protein, it also contains a very small amount of wax substances, carbohydrates, pigments and inorganic components. The specific content varies with species, breeding conditions, inner and outer layers of cocoons, and place of production.

1. Silk Fibroin & Silk Sericin
Although both silk fibroin and sericin are proteins, the percentages of C, H, O, N, and S are different due to the different percentages of amino acids that make up sericin and silk fibroin. Silk can be hydrolyzed into various amino acids by the action of strong acid, alkali and enzyme. After degumming, silk fibroin can be hydrolyzed to determine the amino acid composition. Sericin and silk fibroin are composed of the same 18 amino acids, but their content is different. The non-polar amino acid content in silk fibroin accounts for 3/4 of that in sericin. Non-polar amino acids account for only 1/4. Therefore, sericin is more hydrophilic than silk fibroin. The difference in content also shows that silk fibroin is fibrin and sericin is globulin. 2. Wax
The main components are esters produced by primary alcohols and fatty acids and saturated hydrocarbons.
- Primary alcohol is equivalent to 26 carbon chain beeswax alcohol (C 26 H 53 OH) to 30 carbon chain beeswax alcohol (C 30 H 61 OH);
- Fatty acid is mainly beeswax acid (C 30 H 60 O 2 );
- Saturated hydrocarbons refer to paraffin wax (C 25 H 52 to C 31 H 64 ).
These components are substances that separate the silk fibroin and sericin levels in the liquid silk fibroin, so as not to confuse the two. Therefore, a certain amount of waxy substance can be separated on both sides of the silk fibroin and sericin. The waxy substance in sericin is the most in inner edge sericin.
3. Carbohydrates
Mainly refers to glucose and glucosamine, etc. According to the quantitative study of 18 kinds of cocoon fibers, the average content is 1.64%. The cocoon coat contains 2.4% and silk fibroin contains 0.3%. The presence of sugars is in the form of nitrogen glycosides or glycosides, such as serine anhydride glycosides. The glycoprotein that is combined with glucose and other sugars and inducers has a carbohydrate content of more than 4% is called mucin, and the following is called glycoprotein or glycoconjugate, so sericin can be called glycoprotein. Flavonoid glycosides that bind to pigments are also linked glucose.
4. Pigment
Mainly refers to two categories of flavonoid pigments and carotenoid pigments. The former includes white cocoons, light bamboo blue cocoons, and light green cocoons, while the latter includes yellow cocoons and white cocoons.
- Flavonoids include flavonoids (C 15 H 10 O 2 ), flavonols (C 15 H 10 O 3 ) and flavonoids. The cocoon layer contains about 1% of flavonols. The pigments of white cocoons and green cocoons are the same substance, but the amount that penetrates into the silk glands at the 5th instar is relatively small. The precursors of this pigment are mainly morin (C 15 H 10 O 7 ), and its analogs quercetin (C 15 H 10 O 7 ), isoquercetin (C 12 H 19 O 12 ) and rutin (C 27 H 30 O 16 ).
- Carotenoids include carotenes based carotene or said (C 40 H 56 is ), lutein (C 40 H 56 is O 2 ), and a series of oxide. The red-yellow silk continues to fade in the air or during heating, which is due to continuous oxidation. While oxidizing and fading, the pigment continuously lowers its melting point.
5. Inorganic Substance
The inorganic component ash in the cocoon fiber contains 24 kinds of calcium, magnesium, sodium, potassium, aluminum, iron, silicon, phosphorus, sulfur, manganese, copper, etc., of which calcium, magnesium, sodium, potassium, etc. are more. The total content is about 0.9~1.5%, the outer layer and inner layer of the cocoon are slightly more, and the middle layer is slightly less. Generally, inorganic components are present in sericin and account for about 0.71%, and silk fibroin accounts for about 0.296%. The inorganic elements present in silk fibroin are firmly combined with silk fibroin. For example, if raw silk is boiled with hot water at 120℃, after 5 times of scouring, it still contains 0.23%, which shows that the binding force is very strong.
